A válogatás a Grateful Dead mindegyik stúdióalbumáról a legfontosabbnak számító dalokat tartalmazza, időrendbe állítva.
A számok a banda fejlődését mutatják be – mind zenei, mind a tagok szempontjából- az 1967-es „The Moon”-ig.
A „The Best of The Grateful Dead”-en olyan dalok eredeti verziói is helyet kaptak, melyek később a koncertek nélkülözhetetlen részeivé váltak. Ilyen dal például a „Dark Star”: az itt hallható, ritka kislemez-verzió mindössze két és fél perc hosszú, míg a színpadon sokszor 30 perces, improvizációs showelemmé varázsolta az együttes.
